Use of the micropipette: basic Principles

Micropippettes are basic tools from liquid handling in molecular biology experiments, mainly during master mix preparation, reagent mixtures and PCR running. There are several types, brands and sizes, nevertheless, they share common features. In this video, we can approach to a general use of micropipettes in molecular biology labs.

Running a Endpoint PCR

Polymerase Chain Reaction (PCR) is a worldwide used molecular technique that allows us to amplify DNA (a fragment, a complete region, a complete gene, even a whole chromosome). By using an enzyme, a.k.a. Taq polymerase, and a thermocycler (a device that change the temperature in cycles) we can use a DNA template to create multiple copies of it, detect a specific part or amplify a fragment for sequencing. This video introduces us in a basic protocol for Endpoint PCR. At Laboratory we will replicate this protocol with few modification based on new available devices.

Electrophoresis (Agarose)

Electrophoresis is a molecular technique that allows to separate molecules (nucleic acids, proteins, among others) based on their electric charges in a matrix, in our case a gel matrix. Agarose gel electrophoresis is the most common technique to separate DNA with several advantages

PAGE (PolyAcrylamide Gel Electrophoresis)

PAGE (PolyAcrylamide Gel Electrophoresis), similar to Agarose Gel Electrophoresis, is a molecular technique that allows to separate molecules based on their electric charges in a matrix, in this case a polyacrylamide. PAGE has several advantages mainly related to a high resolution of the bands and a better separation.
